دراسة علاقة القرابة بين فايتوبلازما تورقأزهار السمسم وفايتوبلازما بعض النباتات الاخرى باستخدام تقنية تفاعل البلمرة التسلسلي PCR
Abstract
هدفت الدراسة الى تشخيص فايتوبلازما (Phytoplasma)تورق الازهار (Phyllody) الذي يصيب محصول السمسم Sesame (Sesamumindicum L.) وبعض المحاصيل الاقتصادية المهمة الأخرى وكذلك الادغال المرافقة له في الحقول الزراعية, ومنها ذيل البزونAnnual beard grass (Polypogonmonspeliensis)، الرويطةDarnel (Loliumtemulentum L.)، الطرطيعSuaeda (Schanginiabaccata MQ.T)،الحندقوقMelilotus (Melilotus alba)، الدهنان Purple panic-grass (Echinochloacolonum)، المديد Cynanchum (Cynanchumacutum L.)، الشويل Alkali weed (Cressacretica L.)، زمزومMarvel grass (Dichanthiumannulatum (Forks) Stapf)، البرسيم Alfalfa (Medicago sativa)، القصب Common read (Phragmitescommunis T.)، الشيح Wormwood (Artemisia camperstis L.)، السيسبانSesbania (Sesbaniasesban)، البربينPurslane (Portulacaoleracea)، ومحصول الخيار Cucumber (Cucumissativus) وشجرة فرشة البطل Red bottle brush (Melaleucaviminalis) في مزارع محافظتي الديوانية والنجف الأشرف باستخدام تفاعل البلمرة التسلسلي Polymerase Chain Reaction (PCR) وايجاد علاقة القرابة بين الفايتوبلازما التي تصيب هذه النباتات وفايتوبلازما محصول السمسم.تم تأكيد تشخيص الفايتوبلازما بواسطة تفاعل البلمرة التسلسلي PCR، حيث اثبتت تجربة الكشف عن الحامض النووي DNAلفايتوبلازما تورق الازهار أن نباتات السمسم موضوع الدراسة تحتوي على الحامض النووي DNA وذلك لظهور الحزم الواضحة بحجم (558 bp) في الترحيل الكهربائي لعينة السمسم. كما أثبتت التجربة إلى أن جميع الأدغال المرافقة لمحصول السمسم كانت تحتوي على الحامض النووي DNAلفايتوبلازما تورق الازهار، ما عدى الرويطةDarnel (LoliumtemulentumL.)، المديد Cynanchum (Cynanchumacutum L.) و الشيح Wormwood (Artemisia camperstisL.)، التي اعطت نتائج سالبة.كما إتضحإحتواء بذور السمسم على فايتوبلازما تورق الأزهار والمأخوذة من حاصل حقل التجربة لعام 2015 باستخدام تقنية PCR ولكل من المعاملة والمقارنة.
